Annexin IV (ANX4) belongs to the annexin family of calcium-dependent phospholipid binding proteins. Although their functions are still not clearly defined, several members of the annexin family have been implicated in membrane-related events along exocytotic and endocytotic pathways. ANX4 has 45 to 59% identity with other members of its family and shares a similar size and exon-intron organization. Isolated from human placenta, ANX4 encodes a protein that has possible interactions with ATP, and has in vitro anticoagulant activity and also inhibits phospholipase A2 activity. ANX4 is almost exclusively expressed in epithelial cells. Several transcript variants encoding different isoforms have been found for this gene.

This gene encodes a protein which is a member of the cysteine-aspartic acid protease (caspase) family. Sequential activation of caspases plays a central role in the execution-phase of cell apoptosis. Caspases exist as inactive proenzymes which undergo proteolytic processing at conserved aspartic residues to produce two subunits, large and small, that dimerize to form the active enzyme. This protein cleaves and activates caspases 3 and 7, and the protein itself is processed by caspase 8. Mutations in this gene are associated with type IIA autoimmune lymphoproliferative syndrome, non-Hodgkin lymphoma and gastric cancer. Alternatively spliced transcript variants encoding different isoforms have been described for this gene.

NucView 488 Caspase-3 substrate is a novel, fluorogenic substrate for real-time or endpoint detection of apoptosis in live cells. The non-toxic, no-wash substrate is non-inhibitory to caspase-3 unlike other caspase substrates or FLICA assays permitting multiday analysis. Consisting of a high-affinity DNA dye linked to a DEVD caspase-3 substrate moiety, the initially non-fluorescent, cell permeable substrate is cleaved by caspase-3 in apoptotic cells and emits green fluorescence for the detection of both caspase-3 activity and apopototic nuclear morphology, in single-cells. As apoptotic rates and expression of asociated markers varies between cells, even within the same population, single-cell detection is important. Tribioscience Caspase-1 Activity Fluorometric Assay Kit provides a simple and convenient method for assaying the activity of caspases that recognize the sequence YVAD. The assay is based on the detection of the free AFC fluorescence after cleavage from the labeled substrate YVAD-AFC. The free AFC can be quantified using a fluorometer or a fluorescence microtiter plate reader at Ex400/Em505nm. Comparison of the fluorescence of AFC from an apoptotic sample with an uninduced control allows the determination of the fold increase in caspase-1 activity.
